Biomarkers of Biochemical Toxicity and Oxidative Damage

 

The studies will be focused on:

  • Evaluation of oxidative potential and anticholinesterase activity of selected pesticides in vitro and in vivo.
  • in vitro cell culture models (primary lymphocyte cultures and HepG2 cell lines) will be used to study the oxidative potential of tested pesticides by colorimetric thiobarbituric acid reactive substances (TBARS) assay and antioxidant power by the FRAP test
  • in in vivo studies the effect of pesticide substances on biochemical parameters of interest (activity of acetylcholinesterase (AChE; EC 3.1.1.7) and butyrylcholinesterase (BChE, EC 3.1.1.8) will be evaluated in different tissue samples of rats treated per os with selected doses of pesticides consecutively for 14 days
  • the effects of tested pesticides on oxidative stress will be estimated in blood and tissues of exposed rats by measuring: (1) the concentration of thiobarbituric acid reactive substances (TBARS) as an indicator of lipid peroxidation; (2) activity of antioxidative enzymes such as superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx); (3) the concentration of reduced glutathione (GSH); (4) antioxidant power of plasma and tissue homogenates by FRAP test
